During a team meeting, my supervisor asked me a question and I answered him, but then my colleague called me dirty minded as my answer could be interpreted in a sexual way. Afterwards, my supervisor asked me the same question again with wink when he understood my answer the first time around and my answer was also appropriate. Why did he wink?

We were in discussion with others and he was asking me intentionally a question that involved discussing erectile dysfunction/viagra in front of others - so I refused to talk about it and changed the topic - was he being rude or childish?

The difference is if you were a man he could have been seeking information.  Considering you are a woman it was rude and inappropriate.  What was your response?

What the heck was the context of this question? 

I'm trying to imagine a situation in which a supervisor would even bring this up.

Yes. Asking another male coworker about sexual medication is vastly different than making sexual comments to female coworkers.

As you are a woman there was no reason for him to ask you those questions.

He was trying to make you feel uncomfortable.

His behaviour is inappropriate.

lol, yes that's right. Do you still think it is inappropriate. He asked me the question twice, and he knew that I knew what he was talking about but he wanted to say it out loud.

well we are health care professionals - but he was asking me when he knew I know the answers to his questions. He just wanted me to say it out loud.

Sounds to me like you could both grow up a bit.  He should stop teasing.  And as a health professional, you should know that ED and it's treatment is not something to be embarrassed about.  The words are not embarrassing to say in public.

If he tries it again, just answer dead pan.  If you stop being embarrassed, he'll get bored.

I know that.. but when he asked the question the first time around and it took me few seconds to answer him, he said that I have dirty mind so I laughed... then he asked me the same question again with a wink. I am not the one with immature brain.
